RENAL RENIN CONTENT


1.Take cortical slices and place them in test tubes containing 1 ml of dH20
        (pre-weighed tubes).
2.Store overnight @ -20øC.

3.Remove and let thaw at room temperature.

4.Refreeze (repeat procedure 3 times).

5.Homogenize tissue @ 4øC in phosphate buffer 0.2M; pH:6.5 and add 1.5%
                EDTA_ammonium.
6.Centrifuge and save supernatant.

7.Resuspend pellet and repeat centrifugation.

8.Pool supernatants and adjust with Phosphate buffer. to obtain a final
        solution of 1/500.
9.Measure Renin in supernatant.

10.Save pellet and place in 1 ml of dH2O for protein assay.
